McGahee and Henry cannot be used in the backfield at the same time...

According to Drew Bledsoe this morning there is no way to have the two in all the time. Damian Shelton is a much better blocking back than either one and they need Shelton in the game in many formations. While the two of them in the backfield might "confuse" a team it would leave the Bills with one less blocker.

While the Bills could not have a steady diet of a 2 running back offence, I think there might be a few designed plays where one running back served as a decoy rather than a blocker for the other one.
